Radioimmunoanalytical determination of the specific activity of [125I]L-thyroxine

Abstract

The possibility to determine directly the specific activity of [125I]L-thyroxine by the usual radioimmunoanalytical procedure using the data required for the determination of the calibration graph is demonstrated in this study. This approach, which is from the point of view of the mathematical formalism similar to the double substoichiometric isotopic dilution, can be used - due to its simplicity and sufficient precision in certain cases - for the determination of the specific activity of radioindicators used in the radioimmunoanalysis, particularly for the univalent (from the point of view of immunochemistry) haptenes. The method is limited by the index of heterogeneity of the used antiserum.
