Abstract

Synthesis of oligodeoxyribonucleotides on silica gel support using 5'-O-dimethoxytrityl-(N-acyl)-deoxyribonucleoside 3'-(O-methyl phosphochloridite) intermediates (I) is described. Solutions of these intermediates were prepared before use by mixing of 100 μmol l-1 solution of methyl phosphodichloridite in 1,2-dichloroethane with the same volume of 120 μmol l-1 solution of 5'-O-dimethoxytrityl-(N-acyl)-deoxyribonucleoside in 1,2-dichloroethane containing 1 mol l-1 of sym-collidine. A bench type synthesizer for this methodology is described.
