Theoretical study of the rate determining proton transfer at the mechanism of serine proteases action

Abstract

The proton transfer step at the breakdown of the tetrahedral intermediate of chymotryptic catalysis was studied using the simple quantum-statistical theory of unimolecular proton transfer reactions. The physical basis of the enzymatic high catalytic efficiency is discussed from the view-point of experimental and theoretically obtained kinetic parameters. The experiment is suggested to test the possible importance of vibrational excited states at the enzyme action.
